Brief bio: I joined Prof. Dong Zhang and Mingyu Han’s group at Northwest Agriculture Forestry University since 2015. I work in the Northwest Agriculture Forestry University after finishing postdoctoral experience. My research is mainly focused on apple asexual propagation. In previous studies, we found that inhibition of AR development in apple rootstocks by cytokinin (CK) is based on its suppression of adventitious root (AR) primordium formation, but the molecular mechanism of cytokinin inhibition of AR primordium formation is still unclear. In this study, we found that a low endogenous CK content was beneficial to AR formation in apples. A negative correlation between expression of CK-responsive MdTCP17 and AR formation, while overexpression of MdTCP17 in transgenic apple inhibited AR primordium formation. Further study revealed that an interaction between MdTCP17 and MdWOX11. MdWOX11 has the function of promoting AR primordium formation, MdWOX11 directly binds to the promoter of MdLBD29 and positively regulates its expression. Furthermore, MdTCP17 reduced the binding of MdWOX11 and MdLBD29 promoters, co-expression of MdTCP17 and MdWOX11 reduces MdLBD29 expression. Together, these results explain the function and molecular mechanism of MdTCP17-mediated CK inhibition of AR primordium formation, which could be used to improve apple rootstocks genetically.

个人简历:我从2015开始加入西北农林科技大学园艺学院攻读博士研究生,师从张东教授和韩明玉教授,博士后出站后留校工作。我的研究方向是苹果无性繁殖基础。在前期研究中我们发现细胞分裂素抑制不定根发生源于对不定根原基发生阶段的抑制作用,但是细胞分裂素抑制不定根原基发生的分子机制尚不清楚。在本研究中,我们发现低的内源细胞分裂素含量有利于苹果不定根发生。细胞分裂素响应基因MdTCP17的表达与不定根发生能力呈负相关,而MdTCP17过表达转基因苹果抑制不定根发生。进一步的研究表明,MdTCP17和MdWOX11存在蛋白互作。MdWOX11具有促进不定根发生的功能,MdWOX11直接与MdLBD29的启动子结合并诱导其表达。此外,MdTCP17减弱MdWOX11和MdLBD29启动子的结合,MdTCP17和MdWOX11共表达抑制MdBD29的表达。总之,这些结果解析了MdTCP17介导的细胞分裂素抑制不定根原基形成的分子机制,这可以用于苹果砧木的遗传改良。
