Brief bio: I joined National Resource Center for Chinese Materia Medica, China Academy of Chinese Medical Sciences since 2015. Our group focuses on studying the biosynthetic pathways of plant secondary metabolites. In this work, we verified the functional characterization of diterpene synthases from Leonurus japonicas. Through structure-function analysis identified a critical residue in LjTPS6 (I420) where single site mutations enable specific production of the 13S epimer, elucidate 9,13-epoxy-bridge formation of spiro-labdane diterpenoids. I hope this study can provide a strong foundation for future studies of the intriguing spiro-9,13-epoxy-labdane diterpenoid metabolism.

个人简介:我于2015年加入中国中医科学院中药资源中心开始学习。课题组主要研究方向为植物次生代谢产物生物合成途径解析。在这项工作里,我们对益母草中二萜合酶功能进行研究验证。通过蛋白结构-功能分析,确定了LjTPS6 (I420)一个关键的氨基酸残基,通过该位点突变能够特异性产生13S 空间异构体,阐明了spiro-9,13-epoxy半日花烷型二萜的骨架结构中9,13-epoxy的形成。我们期望这一项研究能够为今后研究spiro-9,13-epoxy-半日花烷型二萜类化合物生物合成研究奠定坚实的基础。
