Brief bio: I have been interested in scientific research since childhood, which led me to choose a major in biology as an undergraduate, and I was further inspired by plant molecular biology. Thereafter, I pursued my master’s degree at the Wuhan Botanical Garden of the Chinese Academy of Sciences, where I conducted research on cytoplasmic male sterility (CMS) in the Brassicaceae crops. To further broaden my scientific horizon, I carried out my PhD research at the Jean-Pierre Bourgin Institute (IJPB) of the French National Academy of Agricultural Sciences under the supervision of Dr. Hakim Mireau and conducted studies on the molecular mechanisms of gene expression in plant organelles and their role in sexual reproduction. In the last few years, we have made exciting progress in the post-transcriptional regulation in plant organelle and the fertility restoration of Ogura-CMS in rapeseed (Wang et al., PNAS 2021; Wang et al., PNAS 2020; Wang et al., NAR 2017; Wang et al., JXB 2018). In the present study, we have identified the molecular function of an unknown PPR protein, MTSF3. MTSF3 exerts a protective activity towards the nad2 exon 3-5 precursor transcript by association to the last nucleotides of its 3’ end and by likely impeding the progression of exoribonucleases. nad2 exon 3-5 associates with another independently transcribed precursor transcripts via a trans-splicing to generate mature nad2 mRNA. MTSF3 carries therefore two intricately connected functions implying the stabilization of both the nad2 exon 3-5 pre-mRNA and the mature nad2 mRNA.

个人简介:我从小就对科学研究充满着向往,于是读本科时候选择了生物学专业,而植物分子生物学进一步启发了我。此后我在中国科学院武汉植物园攻读硕士学位,在汪志伟研究员的指导下开展了十字花科细胞质雄性不育的研究,激发了我对植物核质互作研究的兴趣。为了进一步扩宽科学视野,我来到法国国家农业科学院IJPB研究所进行博士研究,在Hakim Mireau教授指导下开展植物细胞器基因表达分子机制及其在有性繁殖中的作用等相关研究,在植物细胞器基因转录后调控及油菜Ogura-CMS的育性恢复取得了一些令人兴奋的进展 (Wang et al., PNAS 2021; Wang et al., PNAS 2020; Wang et al., NAR 2017; Wang et al., JXB 2018)。 本研究中,我们鉴定到了一个未知PPR蛋白MTSF3的分子功能:MTSF3蛋白结合于线粒体nad2基因的成熟mRNA和一个前体mRNA nad2b的3’ 末端,阻碍核酸外切酶的行进,发挥保护作用。由于前体mRNA nad2b与另一个独立转录的前体mRNA nad2a通过内含子反式剪接,去除内含子从而产生成熟的nad2 mRNA,因而MTSF3携带了两个错综复杂的功能,即同时参与了前体和成熟mRNA 的3’ 端加工事件,并影响了内含子剪接。
